HCM026Dehydrated Culture Medium500g/bottle

Intended Use

For the cultivation of aerobic and anaerobic micro-aerobic and sterility testing of drugs, biological products or other samples.

Principle and Interpretation

Dextrose, pancreatic digest of casein, yeast extract, L-cystine provide the growth factors necessary for bacterial multiplication. L-cystine and sodium thioglycollate allows Clostridium to grow in this medium even under aerobic conditions. Also the small amount of agar used in the medium favors the growth of aerobes as well as anaerobes in the medium, even if sodium thioglycollate is deleted from the medium. Sodium thioglycollate act as a reducing agent and neutralizes the toxic effects of mercurial preservatives and peroxides formed in the medium, thereby promoting anaerobiosis, and making the medium suitable to test materials containing heavy metals. Any increase in the oxygen content is indicated by a colour change of redox indicator, resazurin to red. The small amount of agar helps in maintaining low redox potential for stabilizing the medium.

Formulation

Ingredients/liter
L-Cystine0.5g
Sodium chloride2.5g
Dextrose anhydrous5.0g
Yeast extract5.0g
Pancreatic digest of casein15.0g
Sodium thioglycollate0.5g
Resazurin sodium0.001g
Agar0.75g
pH7.1±0.2 at 25°C

Preparation

Suspend 29.3g in 1 L of distilled or deionized water ,heated to boiling stirring until completely dissolved, dispensing into tubes, autoclave at 121 ℃ for 15min. If more than 1/3 of  the medium is pink, reheat(100℃) it before use to drive off dissolved oxygen. If all the medium is pink, do not use.

Quality Control

Cultural characteristics observed after an incubation at 35-37°C for 24h

Quality control strainsGrowth
Staphylococcus aureus  ATCC6538Good ,broth turbid
Pseudomonas aeruginosa ATCC9027
Clostridium sporogenes  ATCC19404

Storage and Shelf Life

2-30℃,Keep container tightly closed, avoid direct sunlight.

Use before expiry date on the label.

 Precautions

1. When weighing the dehydrated medium, please wear masks to avoid causing respiratory system discomfort

2. Keep container tightly closed after using to prevent clumping.

Waste Disposal

Microbiological contamination was disposed by autoclaving at 121°C for 30 minutes.
